Bacteriophage Pf1 complex viscosity

چکیده

Bacteriophages (phages) are viruses that attack bacteria, causing them to multiply. This requires phage orientation with respect the bacterial receptor, a necessary condition for attachment. Since phages not motile, they rely on their Brownian motion and, specifically, its rotational components reorient. We focus specifically Pf1 (the bacteriophage called pseudomonas Pf1), about which much has been written, though whose diffusivity determined from rheological measurements is known. compare general rigid bead-rod theory intramacromolecular hydrodynamic interactions our new of complex viscosity an aqueous suspension arrive at relaxation time. From this time, we get central transport property reorientation, dimensionless diffusivity, λ0Dr=2.37×10−6, differs within one order magnitude fluorescence microscopy. Characterization of molecular alignment in aqueous suspensions of Pf1 bacteriophage.

The phase diagram of Pf1 solutions has been studied indirectly by observation of 2H quadrupole splittings of the solvent signal and measurement of dipolar couplings in solute macromolecules. At low volume fractions of Pf1 and at high ionic strength, alignment of both the phage and the solute depends strongly on the strength of the magnetic field. Both the theoretical and experimentally determin...

Nuclear spin relaxation of sodium cations in bacteriophage Pf1 solutions.

The nuclear magnetic resonance (NMR) spectra for the I=3/2 23Na cation dissolved into filamentous bacteriophage Pf1 solutions display line splittings and relaxation times consistent with an interaction between the 23Na nuclear quadrupole moment and the electric field gradient produced by the negatively charged Pf1 particles. The 23Na NMR line splittings and relaxation rates corresponding to mag...

Structure of the coat protein in Pf1 bacteriophage determined by solid-state NMR spectroscopy.

The atomic resolution structure of Pf1 coat protein determined by solid-state NMR spectroscopy of magnetically aligned filamentous bacteriophage particles in solution is compared to the structures previously determined by X-ray fiber and neutron diffraction, the structure of its membrane-bound form, and the structure of fd coat protein. These structural comparisons provide insights into several...

Fibres of highly oriented Pf1 bacteriophage produced in a strong magnetic field.

Highly oriented fibres of the cylindrical bacteriophage Pfl have been produced by drying a concentrated solution in a magnetic field of 8 Tesla. Substantial improvement in the alignment of the phage parallel to the fibre axis is demonstrated by X-ray diffraction. This effect is due to the intrinsic diamagnetic aniso· tropy of the viruses and to their co·operative behaviour. The molecular struct...

Ordering of alkali halide salts dissolved in bacteriophage Pf1 solutions: A nuclear magnetic resonance study

The nuclear magnetic resonance ~NMR! spectra of the I53/2 monovalent ions Na, Cl, K, and Br for NaCl and KBr dissolved into filamentous bacteriophage Pf1 solutions display line splittings and shifts consistent with an interaction between the nuclear spin and the electric field gradient produced by the Pf1 particles. The average electric field gradient and thus the spectral splitting for ions in...
